AB While the pathological effects of TSE disease on the CNS are well documented, the molecular events underlying peripheral pathogenesis, particularly in the early stages of disease, are poorly understood. To date the only disease-specific marker for TSE infection is the accumulation of PrPsc but unfortunately detection of PrPsc in easily accessible peripheral tissues is both laborious and technically challenging. In order to aid understanding of the molecular events underlying peripheral pathogenesis and to identify surrogate markers of TSE diseases we have performed a comprehensive analysis of transcripts and proteins in haematopoietic tissue.
Here we report the results of 2-D PAGE analysis of proteins in haematopoietic tissues from control and scrapie infected mice at multiple time points throughout disease. We have utilised the C57/Bl/ME7 i.c. model of TSE disease and have collected spleen, bone marrow and blood at defined time points during this infection. Whole blood was collected and processed into plasma, peripheral blood leucocytes and red cell fraction.
2-D PAGE analysis of blood and bone marrow fractions has enabled us to visualise over 1000 blood proteins and 700 bone marrow proteins. Comparison of protein spot intensities have identified a number of spots representing genes differentially expressed between control and TSE infected animals. As expected, these represented quantitative rather than qualitative differences between control and infected animals. The characterisation of these differentially expressed proteins may help elucidate the role of the hematopoietic system in TSE pathogenesis and may reveal novel surrogate markers of disease.
