AB It is been previously shown that pigs can be susceptible to experimental BSE infection but a Porcine Spongiform Encephalopathy has not been found in pig populations. In order to evaluate the pig/bovine species barrier to prion infection several transgenic mouse lines expressing different levels of the porcine prion protein (poPrPc) were generated. When a high dose BSE inoculum was administered intracerebrally, 14% of inoculated porcine transgenic (poTg) mice became infected and were PrPres positive, as shown by Western blot and IHC analysis. On the other hand, a low dose BSE inoculum could not cross the interspecies barrier in the poTg mice, but infects 42% of control mice. These infectivity analysis indicates the existence of a strong barrier between bovine and pig species. Second passage inoculations were performed with brain preparations from both affected and unaffected poTg mice showing that porcine adapted BSE prions are highly effective infecting poTg mice. These poTg mice seem to be a highly sensitive bioassay for the detection of porcine prions from pig samples suspect of putative prion infection.
