AB It is assumed that the expression level of normal cellular prion protein (PrPc) influences both the rate and the route of transmissible spongiform encephalopathy infection from peripheral entry to the CNS. Animal experiments have shown that the disease is preceded by a period in which the agent accumulates in secondary lymphoid organs (Peyer's patches, lymph nodes, spleen), particularly in association with follicular dendritic cells within the B cell areas.
We have used laser micro-dissection, real time RT-PCR and in situ hybridization on tissue-sections to examine the expression profile of the PrP mRNA in distinct areas of ileal Peyer's patches from normal lambs. Quantification of the PrP mRNA expression in various micro-dissected regions revealed very low expression in the follicles and high expression levels in the inter-follicular area and the muscular layer. These results were supported by in situ hybridization, which showed specific staining in nerve cells in the submucosa and the muscular layer of ileal Peyer's patches. Similar studies on tissue-sections from lambs experimentally infected with scrapie are in progress and results from both studies will be presented.
