AB The Conformation Dependent Immunoassay (CDI) has been demonstrated to provide a highly sensitive approach to the detection of prion disease in animal and man. The InPro CDI(TM) test from Beckman Coulter has been optimized to offer a high level of sensitivity while maintaining the flexibility that can allow the same test to be performed in both high-volume and low-volume BSE testing laboratories. The CDI Low-Volume Kit was developed for the manual testing of a small number of samples while the InPro CDI High-Volume Kit provides the same chemistry in a format which will allow the automated testing of a large number of samples using the Biomek(R) NX Assay Workstation.
Throughout the development of this new assay, we have focused on achieving high levels of sensitivity while simplifying the assay. As with all the InPro CDI assays, this assay relies on a phosphotungstinic acid (PTA) precipitation step to selectively concentrate the diseased form of the prion protein. While using fewer reagents and transfer steps we have increased the sensitivity by performing the immunoassay on a larger portion of the PTA precipitated sample. Sufficient PTA precipitated sample can also be stored for duplicate retest of any initially reactive samples. Duplicate retest of this partially purified extract provides users with a simple approach to confirm the immunoassay portion of the test on those samples which demonstrate an "initial reactant" signal. Detection is performed using the Beckman Coulter DTX Multimode Detector. To guarantee excellent standardization from lab to lab and instrument to instrument, we have established a cut-off value based on femtomoles of europium. By calibrating each instrument against this europium standard, users are assured that equivalent results can be achieved on the same sample in other labs. Sample tracking software using bar coded tubes facilitates the use of this assay in both the low-volume and the high-volume format. For the high volume customers the use of the Biomek NX permits the automated steps to be performed using a single workstation while reducing the space requirements and providing enhanced throughput.
