Bovine Espongiform Encephalopathy and Scrapie are associated to the presence of an unconventional infectious agent called prion. Diagnosis of both diseases is strictly regulated in the EU by several rapid tests and confirmatory tests.
PMCA, previously described for hamster showed the possibility to replicate the misfolded protein in a cell free system. This amplified protein show the same structural an infective properties than original PrPsc.
Here we show the application of PMCA to replicate prion protein from BSE and Scrapie affected animals allowing us to increase the sensibility in the detection of BSE and Scrapie.
Comparison of results obtained with different tests will be shown and molecular characteristics will be analysed in order to identify several uses for this method.
