AB Many studies have been carried out about association between PrP genotype and scrapie within sheep breeds from different countries. In this paper, we report the analysis of prion protein (PrP) polymorphisms in 154 sheep of three African breeds: Bjaad, Sardi and D'man. Haplotypic and genotypic frequencies of known polymorphisms at codons 136, 154 and 171 were determined. The ARQ and resistance-associated ARR haplotypes were found in the three groups. No susceptibility-associated VRQ haplotype was detected in any breed. In addition we identified the previously described polymorphisms at positions 112, 141, 143, 172 and 176. A total of 19 allelic variants and 23 genotypes were found. In D'mans, an allelic frequency of 17.95% for the rare allele 172-D was observed. We also report two new polyporhisms at codon 114 (H->R) in D'man sheep and at codon 146 (N->S) in Bjaad sheep.
