Recently, the SIFT technique (Scanning for Intensely Fluorescent Targets) has been applied for the systematic identification of new anti-prion compounds by high-throughput screening [Bertsch et al. (2005) J. Virol. 79, 7785]. Here, we present the software package SIFTViewer developed by us, which served for rapid visualization and analysis of raw SIFT data and strongly simplified the high throughput screening. It is now in use in follow-up projects.
Bertsch et al. identified the substance class of N'-Benzylidene-Benzohydrazide (NBB) and its structural derivatives to show anti-prion activity in vitro in the SIFT assay and in vivo in a cell culture model of prion diseases. They also studied Structure-Activity Relationships (SAR) of the NBB substance class concerning certain hydroxy groups. Here we extend these SAR studies to halogenic substitutions.
