TI Towards in vivo conditions in biophysical characterization of prions

AB Structure and structural transitions of the prion protein PrP have been studied mostly with recombinant PrP. Those studies have yielded valuable information on basic features of the infection mechanism and pathogenesis and even on applied aspects like diagnosis, decontamination and therapeutic approaches. For studies closer to in vivo conditions, posttranslationally modified PrPc and its natural contact with raft-like lipid membranes were investigated. Conversion of PrPc into PrPsc was studied by seeding the conversion with infectious prion particles, and testing for fibril formation and infectivity. New diagnostic approaches were followed by counting single prion particles, and for heat inactivation complexes of prions and fats and glycerol were studied as they are generated during rendering and oleochemical processes.
