Cell culture models are an essential part of the TSE research for the understanding of cellular and molecular processes leading to the formation and accumulation of the disease inducing agent, PrPres. Cell lines, susceptible for TSE and replicating PrPres, provide a basis for these studies. Unfortunately, only a few of such cell lines exist until now and those are strictly limited to certain, mainly mouse adapted TSE strains.
In our study, we examined a variety of cell lines from different eukaryotic species, tissues and developmental stages for their susceptibility to several TSE strains. We investigated to what extend a successful infection of cell lines is determined by culture conditions (media, split ratios, etc.) or infection protocols. Furthermore, we explored whether the susceptibility of cell lines is strictly related to strains and species barriers.
