AB Cynomolgus monkeys (Macaca fascicularis) are experimentally infectable with bovine spongiform encephalopathy (BSE). BSE-infected monkeys develop the simian form of variant Creutzfeldt-Jakob-Disease (vCJD). Histopathological lesions in the brain resemble those seen in human vCJD including the presence of the so-called florid plaques. In the present study, we present case reports from monkeys that were low-dosed with BSE.
In an European titration study, Cynomolgus monkeys were intracerebrally dosed with log. dilutions of a brain homogenate (bh) from British BSE cattle. All monkeys were female, born in February, 1997 and homozygous for methionine at codon 129 of the prion protein gene. Monkeys were infected with BSE at the age of 3 years.
In an ongoing study, all six high-dosed monkeys (50 mg of a 90% bh) developed the simian form of variant Creutzfeldt-Jakob-Disease (vCJD) within 1000 days post inoculation (dpi), whereas only three out of six monkeys developed the disease when inoculated with 5 mg each. Their incubation period was prolonged (up to 1700 dpi). Clinical symptoms varied among these three cases: only 2/3 showed a wasting syndrome and CNS symptoms ranged from severe ataxia to mild clinical symptoms. The following examinations were done and data will be presented: routine histopathology of hematoxylin-eosin-stained brain tissue sections, immunohistological detection of K-resistant prion protein (PrPres), sensitive detection of amyloid plaques using the fluorochrome BSB, and comparative molecular PrPres typing in different areas of the brain and in peripheral lymphoid tissues using a number of different monoclonal antibodies against PrPc .
