The misfolding and aggregation of normally soluble proteins has emerged as a key feature of several neurodegenerative diseases. In prion diseases, progressive neuronal loss is associated with the accumulation of PrPsc, the misfolded isoform of PrPc.
Previous In vitro studies suggest that cholesterol lowering drugs inhibit the conversion of PrPc to PrPsc and the accumulation of the later, possibly through the disturbance of cholesterol rich membrane domains (lipid rafts). Here we examined the effect of Simvastatin a cholesterol lowering drug, on neurodegeneration in brains of prioninfected mice.
We observed significant delays in disease onset in infected-mice treated with Simvastatin. The delayed disease progression is associated with dramatic neuroprotective effect throughout the infected brains. Further, we found that Simvastatin induced immunomodulatory mechanisms in the infected brains. Our results suggest that Simvastatin delays prion disease in-vivo, through a distinct mechanism of neuroprotection.
