AB Efficient prion infection requires as first step the recognition of the precursor PrPc at specific membrane domains of the cell surface. This population of PrP is extraordinarily complex involving the SecPrP and NtmPrP forms and their processing products, under different ligand-bound states. To decipher this complexity in molecular
terms we have generated major-form expressing cells using PrP mutants which in vitro generate mainly either the NtmPrP or the SecPrP forms. Floatation assays using cold Brij96 extracts show that whereas SecPrP distributes preferentially at the lowest density fractions of a nycodenz gradient, whereas NtmPrP shows a more irregular distribution. Membrane solubilization varying the detergent extraction (type, concentration and temperature) show that both forms significantly differ in the migration properties when analyzed by native gel electrophoresis. The obtained results are interpreted in terms of the role of PrPc heterogeneity in prion amplification.
