TI Mutations at Codons 137, 141, 154 and 176 of the PRNP Gene Significantly Decrease the Susceptibility to Scrapie in ARQ/ARQ Sheep

AB It is commonly considered that only three polymorphic codons, namely 136, 154 and 171 of the PRNP gene are of fundamental importance in moderating resistance/susceptibility to scrapie. As in other breeds, the wild type ARQ sequence of Sarda breed sheep, which is regarded as the most susceptible to scrapie, can contain additional polymorphisms at codons 112, 137, 141, 142, 143, 154 and 176. The association of these polymorphisms with scrapie is in most cases little known, if not unknown. The aim of this work was to investigate the risk of scrapie of the ARQ/ARQ genotype carrying at least one point mutation at codons 112, 137, 141, 142, 154 and 176 in comparison with the ARQ/ARQ without any point mutations. We carried out a study on 256 scrapie-affected sheep and 320 flock-mate negative controls belonging to 24 affected flocks. Twenty ARQ/ARQ experimentally scrapie-inoculated sheep, of which 2 had additional point mutations at codons 176 and 141 and 1 only one point mutation at codon 176, were also included in the study. Logistic regression analysis demonstrated that sheep carrying the ARQ allele with additional mutations had lower risk of becoming sick. ARQ/ARQ genotypes that were detected with heterozygous or homozygous substitutions at codons 137 and 176 had the lowest risk of the disease (Odds Ratio = 0.125 and 0.025). Interestingly, only the 3 experimentally ARQ/ARQ scrapie-inoculated sheep with additional polymorphisms did not develop the disease.
