AB In the last few years, active surveillance system of TSEs in Europe, have been modified in order to detect other strains in Scrapie cases that differs form the typical ones.
An increased number of positive small ruminants to rapid test have been found. During last three years, at least 25 positive goats were analysed in the National Reference Laboratory in Spain and 13 of them were confirmed as atypical samples using discriminatory tests approved in the UE.
Further characterization of its isolates, shows biochemical features in which PrPsc pattern differs from the classical form of the PrPsc in some of its.
In this study, we analyzed molecular and histopathological profile from these atypical goat cases in Spain and geographical distribution compared with classical cases from ovine and caprine.
Comparison of results based on glycoform ratio, molecular mass and monoclonal antibody recognition of PrPsc carried out, together histological studies allow us to discard BSE presence in those animals.
The hole ORF of the PRNP gene in all the goats were sequenced in order to determine polymorphisms associated with resistance to scrapie in goats like the ones described previously (Vaccari et al 2006) in positions 154 and 222 and possible new mutations detected.
